What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests GI-101/GI-101A alone or with pembrolizumab or lenvatinib in adults with solid tumors that are advanced or have spread. It looks at safety, side effects, how the body handles the drugs, and how well they work.
The purpose of this study is to evaluate the safety, tolerability, pharmacokinetics, and therapeutic activity of GI-101/GI-101A as a single agent or in combination with pembrolizumab or lenvatinib over a range of advanced and/or metastatic solid tumors.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Age 18 or older, or 19 or older if local rules require it.
- Organs and bone marrow must work well enough to meet study rules.
- Cancer must be measurable using the study’s rules.
- Must be fully active or able to do light work, with limits only on hard physical activity.
- Side effects from past cancer treatment or surgery must be mild or gone. Hair loss and moderate nerve damage in the hands or feet are allowed.
- People with HIV must take HIV medicine and have well-controlled HIV.
Who can't join
- Known active cancer spread to the brain or spinal cord, or cancer in the tissue covering them.
- Another active cancer.
- Current or past hepatitis B or active hepatitis C (liver infections); current or past active tuberculosis; active or uncontrolled infections; or a severe infection within 4 weeks before treatment.
- Long-term liver disease or liver scarring, except in people whose cancer has spread to the liver.
- An active autoimmune disease, where the immune system attacks the body, that needed whole-body treatment in the past 2 years.
- An immune system disorder that weakens infection defenses, or long-term whole-body steroids or other immune-weakening medicines within 2 weeks before treatment.
- Past immune treatments that work like GI-101; whole-body cancer treatment, including study drugs, within 4 weeks; or radiation within 2 weeks, except limited-area radiation to ease symptoms.
- A live, weakened vaccine within 4 weeks before treatment, or a known allergy to any ingredient in GI-101/GI-101A, pembrolizumab, or lenvatinib.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Key Inclusion Criteria: * Males and females aged ≥ 18 years (or ≥ 19 years according to local regulatory guidelines) at the time of screening. * Has adequate organ and marrow function as defined in protocol. * Measurable disease as per RECIST v1.1. * ECOG performance status 0-1. * Adverse events related to any prior chemotherapy, radiotherapy, immunotherapy, other prior systemic anti-cancer therapy, or surgery must have resolved to Grade ≤1, except alopecia and Grade 2 peripheral neuropathy. * HIV infected patients must be on anti-retroviral therapy (ART) and have a well-controlled HIV infection/disease as defined in protocol. Key Exclusion Criteria: * Has known active CNS metastases and/or carcinomatous meningitis. * An active second malignancy * Has active or a known history of Hepatitis B or known active Hepatitis C virus infection. * Has active tuberculosis or has a known history of active tuberculosis * Active or uncontrolled infections, or severe infection within 4 weeks before study treatment administration. * History of chronic liver disease or evidence of hepatic cirrhosis, except patients with liver metastasis. * Has an active autoimmune disease that has required systemic treatment in past 2 years. * Previous immunotherapies related to mode of action of GI-101. * Has a diagnosis of immunodeficiency or is receiving chronic systemic steroid therapy or any other form of immunosuppressive medications within 2 weeks prior to Cycle 1 Day 1. * Administration of prior systemic anti-cancer therapy including investigational agents within 4 weeks prior to treatment. * Radiotherapy within the last 2 weeks before start of study treatment administration, with exception of limited field palliative radiotherapy * Administration of a live, attenuated vaccine within 4 weeks before Cycle 1 Day 1. * Known hypersensitivity to any of the components of the drug products and/or excipients of GI-101/GI-101A, pembrolizumab or lenvatinib. Other protocol defined inclusion exclusion criteria may apply. Cancer type and part-specific inclusion criteria are described in the study protocol.
