What to expect

No one can promise a trial will help you personally — that is what the research is trying to find out.

What participation involves

  • Time periods mentioned: 1 year, 10 years

Only what the study's summary and detailed description on ClinicalTrials.gov say. Time periods may describe the whole study or a part of it — ask the study team what applies to you.

Show the study's own words
  • "It is recommended that patients be followed every 4 months for 1 year, every 6 months for 1 year and then yearly until the patient is in remission for 10 years and is at least 18 years old."
Compensation
Not stated
  • Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
  • Participants are monitored closely by the research team throughout the study.
  • Some participants may receive access to an investigational treatment, which is still being studied.
  • Your participation helps researchers learn whether this approach could help future patients.

About this study

In plain language

This study tests whether adding dasatinib or venetoclax to chemotherapy improves treatment results. It is for children and teens ages 1 to 18 with newly diagnosed T-cell leukemia or lymphoma, or mixed phenotype leukemia or lymphoma (cancer with more than one cell type).

This is a clinical trial testing whether the addition of one of two chemotherapy agents, dasatinib or venetoclax, can improve outcomes for children and young adults with newly diagnosed T-cell acute lymphoblastic leukemia and lymphoma or mixed phenotype acute leukemia. Primary Objective * To evaluate if the end of induction MRD-negative rate is higher in patients with T-ALL treated with dasatinib compared to similar patients treated with 4-drug induction on AALL1231. * To evaluate if the end of induction MRD-negative rate is higher in patients with ETP or near-ETP ALL treated with venetoclax compared to similar patients treated with 4-drug induction on AALL1231. Secondary Objectives * To assess the event free and overall survival of patients treated with this therapy. * To compare grade 4 toxicities, event-free survival (EFS) and overall survival (OS) of patients treated with this therapy in induction and reinduction to toxicities of similar patients treated on TOT17.

Compensation is read from the study's own description and may be missing — always confirm with the study team.

Eligibility

Who can join

  • Must be enrolled in INITIALL and be age 1 to 18.99 when joining it.
  • Have T-cell acute lymphoblastic leukemia or lymphoma, or mixed phenotype acute leukemia or lymphoma.
  • Have had no chemotherapy except treatment given or allowed by INITIALL.
  • Have completed no more than 3 days of chemotherapy on INITIALL.
  • Have direct bilirubin, a liver blood test, no higher than 1.5 times the normal upper limit for age.
  • Have ALT, another liver blood test, no higher than 5 times the normal upper limit for age.
  • Meet kidney test limits: a kidney filtering rate of at least 50, or a creatinine blood level within the study's limits for age and sex.

Who can't join

  • Cannot or do not want to give the required consent or agreement to join.
  • For people with T-cell lymphoma only: have nerve damage rated above grade 2 when joining.
  • Have trouble absorbing medicines or another condition that prevents taking medicine by mouth.
  • Have known HIV infection, a positive hepatitis B surface antigen test, or a positive hepatitis C antibody test.
  • Are pregnant or breastfeeding.
  • Could have children but will not use highly effective birth control during treatment and for 90 days after.
  • Have taken certain medicines or supplements that affect how the body breaks down drugs within 7 days before treatment. These include rifampin, carbamazepine, phenytoin, and St. John's wort.
  • Have eaten grapefruit, grapefruit products, Seville oranges, or starfruit within 3 days before treatment.
