About this study
In plain language
This study tests whether kidney problems change how much GL0034 stays in the blood and checks its safety. It is for adults ages 18 to 80 with normal kidney function or moderate to severe kidney problems, including some people on dialysis. Each person gets one shot under the skin and has blood tests.
This is an open-label, single-dose, Phase 1 clinical study designed to evaluate the effect of renal impairment on the pharmacokinetics (PK) of GL0034, a long-acting GLP-1 receptor agonist. Approximately 40 adult participants will be enrolled across four groups: normal renal function, moderate renal impairment, severe renal impairment without dialysis, and severe renal impairment with dialysis. Each participant will receive a single subcutaneous dose of GL0034. Blood samples will be collected for PK analysis. Secondary objectives include assessing safety and tolerability. The study will help determine whether renal impairment affects GL0034 exposure and inform dosing recommendations for patients with compromised renal function.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Be 18 to 80 years old, agree to join in writing, and be able to follow study visits and instructions.
- Have a kidney filtering rate (eGFR) of at least 90, 30–59, or below 30 mL/min. People below 30 may be off dialysis or get hemodialysis, which uses a machine to clean the blood at set times.
- Have a body mass index (BMI), a measure of weight compared with height, from 18 to 42.
- Women who can become pregnant and are sexually active must follow birth control rules from 4 weeks before the shot until 30 days after it.
- Men must follow birth control rules from the start of the study until at least 30 days after the shot. They must not donate sperm during the study or for at least 30 days after it ends.
- People with kidney problems must have had them for at least 3 months, with no major change in the past 30 days, and otherwise be in good health.
- People with kidney problems must have resting blood pressure with a top number of 90–160 and a bottom number no higher than 110. Their pulse must be no higher than 110 beats per minute.
- People taking medicines or treatments for conditions linked to kidney problems must keep them stable for at least 4 weeks before the shot and during the study.
Who can't join
- Use continuous blood-filtering treatment or peritoneal dialysis, which cleans blood through the lining of the belly.
- Have calcitonin, a hormone, at 50 ng/L or higher; a personal or family history of medullary thyroid cancer or MEN type 2, an inherited condition that causes tumors; or past pancreatitis, swelling of the pancreas.
- Take medicines such as digoxin or warfarin that need very careful dosing, or have an allergy to the study drug or its ingredients.
- Have had major depression in the past 2 years, any suicide attempt, or alcohol or drug abuse in the past 2 years. A positive urine drug test may also exclude you, unless a prescribed medicine caused it; this test is waived for people with kidney failure who make no urine.
- Have had a heart attack, unstable chest pain, or a stroke from blocked blood flow in the past 6 months; a serious abnormal heart tracing; active cancer; or another illness that makes joining unsafe. Some past, treated cancers are allowed.
- Have certain abnormal liver, blood count, or blood-clotting tests, or a bleeding problem that could cause too much bleeding from the shot.
- Test positive for HIV, hepatitis B, or hepatitis C. People with hepatitis C antibodies may join if a follow-up test finds no virus.
- Have recently joined another treatment study or received an experimental treatment; be placed in an institution by an official or court order; or have ties to study staff or the sponsor that could create a conflict of interest.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria: 1. Participant is willing and able to sign a written or electronic informed consent. 2. Participant is an adult male or female, aged between 18 and 80 years of age, inclusive, at the time of screening. 3. Examples of medically highly effective forms of birth control are: 1. Confirmed infertility due to surgical procedure or post-menopausal (cessation of menses for at least 12 months prior to Screening). 2. Confirmed infertility of sexual partner or partner of the same sex. 3. Hormonal contraceptive (oral, combined, patch, vaginal ring, injectable, implant) in females. 4. Double-barrier method (any combination of physical and chemical methods). 5. Intrauterine device with a failure rate less than 1% per year. Male participants must: 6. Agree to use, with their partners, one of the highly effective contraceptive methods, from Baseline until at least 30 days following last dose of trial drug. 7. Refrain from donating sperm during the trial and for at least 30 days after the end of the trial. 6. Participant is willing and able to comply with the study protocol, visit schedule, and other study-related instructions and procedures. Participants with Renal Impairment 7. The participant must have no clinically significant change in disease status within the last 30 days before screening, as documented by the participant's recent medical history. 8. The participant must have a condition consistent with renal impairment, for at least 3 months, and associated symptoms, but otherwise be determined to be in good health in the opinion of the Investigator. 9. The participant must have blood pressure (after the participant is supine for at least 5 minutes) between 90 and 160 mmHg systolic, inclusive, and not higher than 110 mmHg diastolic. If blood pressure is out of range, up to 2 repeated assessments are permitted at Screening and check-in. Pulse should not be higher than 110 beats per minute (bpm). 10.
