What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests how well different doses of frevecitinib (KN-002) work and how safe they are over 12 weeks. It is for people with severe asthma that is not controlled by medium- to high-dose inhaled steroids plus a long-acting medicine that opens the airways.
A Phase 2 Dose Ranging Study to Evaluate the Efficacy and Safety of Frevecitinib (KN-002) Over a 12-Week Treatment Period in Patients With Severe Asthma Not Controlled With Medium to High Dose ICS/LABA
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Understand and sign the consent form, with a witness, and agree to follow study rules.
- Have a body mass index (BMI, a measure of weight compared with height) of 18–40 and weigh at least 40 kg (about 88 pounds).
- Have doctor-diagnosed asthma for at least 12 months.
- Have used medium- or high-dose inhaled steroids plus a long-acting airway-opening medicine for at least 6 months. Steroid and other controller doses must be stable for at least 4 weeks before screening and during the screening period.
- Meet breathing test limits: blow out 40%–80% of the expected air in one second before an airway-opening medicine. After the medicine, this amount must improve by at least 12% and 200 mL.
- Score at least 1.5 on an asthma control survey at both screening visits. Have had at least one asthma flare-up in the past 12 months while using the required inhalers.
- Use inhalers and breathing test tools correctly. Complete at least 80% of required electronic reports during the last 14 days of screening.
- If able to become pregnant and sexually active with a male partner who has not been sterilized, use highly effective birth control from consent through 16 weeks after the last study dose.
Who can't join
- Currently smoke, or have smoked at least 10 pack-years, such as one pack a day for 10 years.
- Have current angina (heart-related chest pain); a heart attack, stroke, or mini-stroke in the past year; past blood clots or certain conditions that raise clot risk; or a heart rhythm test result called QTcF above 470 milliseconds.
- Have another lung disease that could affect the study, unsafe exam or lab findings, active liver disease or certain liver tests above twice normal, or any history of cancer.
- Have a breathing infection that has not cleared by screening or occurs during screening or on Day 1. Have a significant infection or take whole-body antibiotics, parasite medicines, or antiviral medicines on Day 1. Certain TB, hepatitis B or C, or HIV histories or test results, or use of HIV medicines, also exclude you.
- Have an allergy to a study drug ingredient or another allergy that makes joining unsafe. Use certain medicines within the study's time limits, including JAK inhibitors, immune-suppressing drugs, experimental drugs, steroid pills or short courses of whole-body steroids, or injected antibody treatments.
- Are pregnant or breastfeeding, or have had long-term alcohol or drug abuse within the past 12 months.
- Plan surgery during the study that needs general anesthesia or a hospital stay longer than one day.
- Received a live or weakened-live vaccine within 15 days before screening, or have had bronchial thermoplasty, a heat treatment for asthma.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria: Capable of understanding the written informed consent, provides signed and witnessed written informed consent prior to any study-related procedures, and agrees to comply with protocol requirements. Body mass index between 18 to 40 kg/m2 and weight ≥40 kg at screening. Documented physician-diagnosed asthma for at least 12 months prior to screening. Has received a physician-prescribed asthma controller regimen with medium dose or high dose ICS plus LABA, with or without additional controller medications for at least 6 months prior to screening and the dose of ICS and additional controller(s) must be stable for at least 4 weeks prior to screening and throughout the screening/run-in period. Has a pre-bronchodilator FEV1 value of ≥40% and ≤80%, predicted, at screening and at Day 1. Has a post-bronchodilator reversibility of FEV1 ≥12% and ≥200 mL documented during screening (15 to 30 min after administration of 4 puffs of albuterol/salbutamol). Has an ACQ-6 score of ≥1.5 during screening (ie, Visit 1 and Visit 2). Has a documented history of at least 1 asthma exacerbation in the 12 months prior to the screening visit, while using medium to high dose ICS/LABA therapy Acceptable inhaler, peak flow meter, and spirometry techniques during the screening/run-in period. ≥80% compliance with required use of the ePRO device within the last 14 days of the screening/run-in period. Females of childbearing potential who are sexually active with a non-sterilized male partner must use a highly effective method of contraception from the time informed consent is obtained and must agree to continue using such precautions throughout the study and continue using such precautions for 16 weeks after the final dose of study treatment. Exclusion Criteria: Current smokers or participants with a smoking history of ≥10 pack years Participants with a current history of angina or history of myocardial infarction, stroke, or TIAs within the past 12 months from screening are disallowed. Participants with a history of pulmonary embolic or thrombotic events, or genetic or autoimmune (eg anti-phospholipid syndrome) predisposition for thrombosis are disallowed.
