A Study to Evaluate the Drug Levels, Absolute Bioavailability, Safety, Tolerability, and Immunogenicity of Single-Dose of BMS-986446 in Healthy Adults After Single-dose Administration, and Participants With Early Alzheimer's Disease After Multiple Dose Administration
A Phase 1, Open-label, Multi-part Study to Evaluate Pharmacokinetics, Absolute Bioavailability, Safety, Tolerability, and Immunogenicity of Single-Dose of BMS-986446 Following Intravenous and Subcutaneous Administrations in Healthy Adults and Multiple Doses of BMS-986446 in Participants With Early Alzheimer's Disease.

What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests BMS-986446 in healthy adults after one dose and in people with early Alzheimer's disease after several doses. It checks how much drug reaches the blood, its safety and side effects, and whether the body's immune system reacts to it.
The purpose of this study is to evaluate the drug levels, absolute bioavailability, safety, tolerability, and immunogenicity of single-Dose of BMS-986446 in healthy adults after single-dose administration, and participants with early Alzheimer's Disease after multiple dose administration
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Have a body mass index (BMI), a measure of weight compared with height, from 18 to 35.
- For Parts A and B: Be healthy based on health history, exams, heart and blood tests, and a check for suicide risk.
- For Part C: Have mild cognitive impairment (mild problems with memory or thinking) or mild Alzheimer's dementia that meets the study's diagnosis rules.
- For Part C: Score from 20 to 28 on the MMSE, a test of memory and thinking.
- For Part C: Have a positive blood test for pTau217, a marker linked to Alzheimer's disease.
Who can't join
- For Parts A and B: Have a history of major health problems involving digestion, kidneys, liver, lungs, brain, mental health, heart, hormones, immune system, blood, or how the body uses food for energy. Cancer, severe liver scarring, and ongoing allergies needing treatment also exclude you.
- For Parts A and B: Have donated or lost 500 mL of blood or more in the 60 days before receiving the study drug.
- For Part C: Have another brain or nerve condition that the study doctor thinks may affect memory or thinking. Examples include Parkinson's disease, other types of dementia, seizures, brain tumors, multiple sclerosis, or long COVID.
- For Part C: Have a current main mental health diagnosis other than Alzheimer's disease, such as major depression, schizoaffective disorder, or bipolar disorder, or symptoms such as seeing things that are not there or holding false beliefs.
- Other study rules for who can and cannot join also apply.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria * Participants must have a BMI of 18.0 to 35.0 kg/m2. * For Parts A and B: Participants must be healthy as determined by medical history, Physical Examination (PE), neurological examination, vital signs, 12-lead ECG, Columbia Suicide-Severity Rating Scale (C-SSRS), and clinical laboratory evaluations. * For Part C: Participants must meet diagnostic criteria for MCI or mild AD dementia, consistent with the National Institute on Aging and the Alzheimer's Association (NIA-AA) diagnostic criteria. * For Part C: Participants must have an Mini Mental State Examination (MMSE) score of ≥ 20 to 28 (inclusive). * For Part C: Participants must have evidence of positive plasma pTau217. Exclusion Criteria * For Parts A and B: Participants must not have a general history of any clinically significant gastrointestinal, renal, hepatic, broncho-pulmonary, neurological, psychiatric, cardiovascular, endocrinological, immune-mediated disorder, hematological, ongoing allergic disorder requiring treatment, metabolic disorder, cancer, or cirrhosis. * For Parts A and B: Participants must not have donated or lost 500 mL blood or more within 60 days prior to study intervention administration. * For Part C: Participants must not have a neurological condition that in the opinion of the investigator may be contributing to cognitive impairment aside from the AD diagnosis, including but not limited to: Parkinson's disease, vascular dementia, dementia with Lewy bodies, frontotemporal dementia, Huntington's disease, normal pressure hydrocephalus, brain tumor, progressive supranuclear palsy, seizure disorder, subdural hematoma, multiple sclerosis, long COVID, or baseline intellectual disability. * For Part C: Participants must not have any current primary psychiatric diagnosis (eg, major depression, schizoaffective disorder or bipolar disorder) other than AD or symptoms (eg, hallucination or delusions). * Other protocol-defined Inclusion/Exclusion criteria apply.
