About this study
In plain language
This study tests how safe Pluvicto is and how well it works in African American men with prostate cancer that has spread and keeps growing despite treatment to lower testosterone. Pluvicto finds a protein on prostate cancer cells and uses radiation to kill them.
This phase II trial tests how well Pluvicto works in treating African American patients with prostate cancer that has spread from where it first started (primary site) to other places in the body (metastatic) and that has continued to grow and spread despite the surgical removal of the testes or medical intervention to block androgen production (castration-resistant). Pluvicto is a targeted radioligand therapy. It works like a guided missile, combining tumor seeking molecules with a radioactive isotope. It circulates through the bloodstream, binds to a protein on prostate cancer cells called prostate specific membrane antigen, and destroys them using radiation. Pluvicto is approved by the Food and Drug Administration for the treatment of metastatic castration-resistant prostate cancer. This trial may help researchers determine the safety and efficacy of Pluvicto specifically in African American patients with metastatic castration-resistant prostate cancer.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Be age 18 or older and identify as African American.
- Understand and sign the consent form, and follow study and radiation safety rules.
- Be fully active or able to do light work despite some limits on hard physical activity.
- Have prostate cancer confirmed by tissue or cell tests, with a positive PSMA scan. PSMA is a protein on prostate cancer cells.
- Have cancer that is getting worse despite very low testosterone, with spread shown on a recent CT, MRI, or bone scan.
- Have cancer that got worse only once on a newer hormone-blocking drug, which must be your most recent treatment. Your doctor must consider a switch to another such drug suitable.
- Have no more than moderate lasting side effects from earlier treatments, except hair loss.
- Meet the study’s blood count, liver, kidney, and blood protein test limits.
Who can't join
- Have had PSMA-targeted radiation treatment, certain other radiation treatments within 6 months, or a study drug within 28 days.
- Have had chemotherapy or certain immune or other cancer treatments, or are receiving treatments not allowed by the study. Some past taxane chemotherapy and sipuleucel-T are allowed.
- Have an allergy to study treatments or similar drugs, or receive blood transfusions or blood-cell growth drugs only to meet study entry rules.
- Have unstable cancer spread to the brain or spinal cord, symptoms from that spread, or need steroids for it. Also excluded: pressure or threatened pressure on the spinal cord, or a bone scan pattern called a superscan.
- Have certain serious heart problems, uncontrolled infection, active hepatitis B or C, or another serious illness that could prevent participation. Current COVID-19 requires full recovery.
- Have another cancer that may shorten life or affect study test results. Some fully treated past cancers, skin cancers, and shallow bladder cancers are allowed.
- Are unwilling to use condoms during sex or avoid donating sperm during treatment and for 14 weeks after treatment ends.
- Have blocked urine flow or urine leaks that cannot be controlled with care.
Simplified for readability. The official criteria below are what the study team uses.
