What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests the safety of AZD2284, AZD2287, and AZD2275 and how well people tolerate them. It is for people with prostate cancer that has spread and is resistant to treatment that lowers testosterone.
The main purpose of the study is to assess the safety and tolerability of AZD2284, AZD2287, and AZD2275.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- You can do all or most daily activities (ECOG score of 0 or 1), and your organs work well enough.
- A tissue test shows prostate adenocarcinoma, a type of prostate cancer. Doctors must not strongly suspect that most of the cancer is a neuroendocrine type.
- You have had both testicles removed, are taking treatment to lower testosterone, or both. Your testosterone must be below 50 ng/dL.
- A CT, MRI, or bone scan taken within 28 days before the first study dose shows cancer has spread. Cancer that has spread only to bones can qualify.
- For Part A, the cancer may be stable or getting worse. For Part B, it must be getting worse based on PSA blood tests, soft-tissue growth, or bone disease.
- For the Part B dose-increase group, you must have had at least 2 treatments that work throughout the body for this cancer. For the dose-expansion group, you need at least 1. Both groups require a drug that blocks male hormone signals (ARPI).
- Part B also requires certain past treatments based on tumor tests: a PARP inhibitor for a BRCA gene change, and a checkpoint inhibitor for MSI-H, dMMR, or TMB of at least 10. These tests measure DNA repair problems or gene changes. In dose expansion, the BRCA treatment follows local practice and may be waived if the study doctor says you cannot receive it.
- For Part B dose expansion, you must not have had cell-killing chemotherapy for hormone-resistant cancer. Taxane chemotherapy for hormone-sensitive cancer is allowed if the last cycle started over 12 months ago. Past PSMA-targeted radiation drugs, radium-223, or other radiation may be allowed if you meet the other rules.
Who can't join
- You received a radioactive drug within 6 weeks before the first study dose, or cell-killing chemotherapy or an experimental treatment too recently. For these other treatments, the limit is 28 days or 5 drug half-lives, whichever is shorter; a half-life is the time for the drug level to fall by half.
- You had radiation within 28 days before the first dose and still have radiation side effects that are more than mild.
- Side effects from past treatment have not improved to mild or less.
- You have a serious or uncontrolled illness unrelated to cancer, or a social situation that would keep you from following study rules.
- You have a known or suspected allergy to a study drug or its ingredients, or another medical reason you cannot take it.
- You have a medically important amount of protein in your urine.
- A bone or PSMA scan shows very strong, widespread uptake of the scan tracer in your bones, called a superscan.
- You regularly take corticosteroid medicine at a daily dose greater than 10 mg of prednisone or an equal dose of another steroid.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria: * eria: * • Eastern Cooperative Oncology Group (ECOG) Performance Status of 0 or 1. * • Histologically confirmed diagnosis of adenocarcinoma of the prostate without strong clinical suspicion of majority neuroendocrine differentiation. * • Must have had prior bilateral orchiectomy and/or ongoing androgen-deprivation therapy and a castrate level of serum/plasma testosterone (\< 50 ng/dL or \< 1.7 nmol/L). * • At least one metastatic lesion present on baseline Computed Tomography (CT), Magnetic Resonance Imaging (MRI), or bone scan obtained ≤ 28 days prior to the first dose of Investigational Medicinal Product (IMP). Participants may have non-measurable lesions including bone only metastases. * • Adequate organ function * • Part A only: Metastatic prostate cancer considered to be stable or progressing metastatic castration resistant prostate cancer (mCRPC). * • Part B only: Progressing mCRPC defined as meeting at least one of following documented criteria - * a)Serum/plasma PSA progression * b)Soft-tissue progression * c)Progression of bone disease * • Part B Dose Escalation: Previously treated with at least 2 prior lines of systemic anti-cancer therapy for mCRPC. Prior lines must include: * a)At least 1 androgen receptor pathway inhibitor (ARPI) * b)A poly (adp-ribose) polymerase (PARP) inhibitor for participants with known BRCA mutation * c)A checkpoint inhibitor for participants with known microsatellite instability-high (MSI-H), deficient mismatch pair (dMMR), or tumor mutational burden (TMB) ≥ 10 mut/Mb * • Part B Dose Expansion: Previously treated with at least 1 prior line of systemic anti-cancer therapy for mCRPC. Prior lines must include: * (a)At least 1 ARPI * (b)A PARP inhibitor for participants with known BRCA mutation per local practice, unless ineligible per Investigator decision. * (c)A checkpoint inhibitor for participants with known MSI-H, dMMR, or TMB ≥ 10 mut/Mb. * (d)No previous cytotoxic chemotherapy for CRPC. Taxanes for metastatic hormone sensitive prostate cancer (mHSPC) is acceptable if the last cycle Day 1 was \> 12 months before first study treatment. * (e)Previous treatment with prostate specific membrane antigen radioligand therapy (PSMA-RLT) or Radium-223 is allowed but not required. Participants who have had prior radiation therapy, including therapeutic radiopharmaceuticals, external bean radiation therapy (EBRT), and/or brachytherapy are eligible, subject to satisfying all other inclusion/exclusion criteria. Therapeutic radiopharmaceuticals will be considered a prior line of systemic therapy. * Main Exclusion Criteria: * • Treatment with any radiopharmaceutical within 6 weeks of the first dose of Investigational Medicinal Product (IMP). * • Radiation therapy (RT) or external beam radiation therapy (EBRT) within 28 days prior to the first dose and all RT-related events have not recovered to Grade ≤ 1. * • Administration of any systemic cytotoxic or investigational therapy ≤ 28 days of the first dose of IMP or 5 half-lives, whichever is shorter. * • All prior treatment-related adverse events must have resolved to Grade ≤ 1. * • Concurrent severe and/or uncontrolled illness not related to cancer and/or social situation that would limit compliance with study requirements. * • Known or suspected allergies or contraindications to any of the investigational drugs or any component of the investigational drug formulation. * • Clinically relevant proteinuria * • Diffuse and intense osseous radiotracer uptake on bone scintigraphy or PSMA imaging characteristic of a superscan. * • Chronic corticosteroid use greater than 10 mg prednisone equivalent daily.
