What to expect

No one can promise a trial will help you personally — that is what the research is trying to find out.

What participation involves

Not stated in the study description — ask the study team about visits and procedures.

Compensation
Not stated
  • Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
  • Participants are monitored closely by the research team throughout the study.
  • Some participants may receive access to an investigational treatment, which is still being studied.
  • Your participation helps researchers learn whether this approach could help future patients.

About this study

In plain language

This study tests Risvutatug rezetecan (Ris-Rez, also called GSK5764227), alone or with other treatments, for advanced digestive system cancers that cannot be removed by surgery or have spread. It checks how well the medicine works, its safety, and how the body handles it in people who have already had cancer treatment.

This study will check how well a new medicine, Risvutatug rezetecan, (Ris-Rez, also known as GSK5764227) works, how safe it is and how the body handles it in participants all around the world with advanced inoperable or metastatic gastrointestinal cancer who have previously received treatment.

Compensation is read from the study's own description and may be missing — always confirm with the study team.

Eligibility

Who can join

  • Be at least 18 years old, or the legal age to give consent where the study takes place.
  • Have colon or rectal adenocarcinoma, a cancer that starts in gland cells, confirmed by a tissue test. It must be advanced or have spread and cannot be removed by surgery.
  • Or have pancreatic adenocarcinoma confirmed by a tissue or cell test. It must be advanced and unable to be removed by surgery, or have spread.
  • Be willing to use suitable birth control.
  • Be able to sign a consent form and follow the study rules.
  • Be fully active or able to walk and do light work, even if heavy activity is limited.
  • Have organs that work well enough for the study.

Who can't join

  • Have another cancer that grew or needed treatment in the past 24 months. Exceptions include certain skin cancers or early cancers removed with no signs left.
  • Had major surgery within 28 days before study group assignment or the first dose, depending on the group; or ever had a bone marrow or solid organ transplant.
  • Have severe, uncontrolled, or active heart or blood vessel problems, or serious or poorly controlled high blood pressure.
  • Had major bleeding symptoms or a strong tendency to bleed within 1 month before the first dose.
  • Have cancer that spread to the brain or central nervous system and is untreated or has gotten worse.
  • Have lung scarring or inflammation not caused by infection, had it before and needed high-dose steroids, or may have it and scans cannot rule it out.
  • Still have side effects from past treatment that are more than mild and have not returned to their earlier level; or previously had an antibody-linked cancer drug carrying a topoisomerase-1 blocker.
  • Are pregnant or breastfeeding.

Simplified for readability. The official criteria below are what the study team uses.

Official eligibility criteria

Inclusion criteria: Participants are eligible to be included in the study only if all of the following criteria apply: All cohorts * Is at least 18 years of or the legal age of consent in the jurisdiction in which the study is taking place at the time of signing the informed consent form (ICF). CRC Cohort * Has histologically confirmed unresectable, locally advanced or unresectable metastatic adenocarcinoma of the colon or rectum (histology defined by World Health Organization (WHO) classification). PDAC Cohort * Has histologically or cytologically confirmed unresectable, locally advanced or metastatic adenocarcinoma of the pancreas (histology defined by WHO classification). All Cohorts * Is willing to use adequate contraception. * Is capable of giving signed informed consent, including compliance with the requirements and restrictions listed in the ICF and in the protocol. * Has an ECOG performance status of 0 or 1. * Has adequate organ function. Exclusion criteria: Participants are excluded from the study if any of the following criteria apply: * Has a malignancy (except disease under study) that has progressed or required active treatment within the past 24 months except for basal cell or squamous cell carcinomas of the skin or in-situ carcinomas \[e.g., breast, cervix, bladder\] that have been resected with no evidence of disease. * Has had any major surgery within 28 days prior to randomization or first dose of study intervention, depending on sub-cohort/cohort assignment * Has any history of prior allogenic or autologous bone marrow transplant or other solid organ transplant. * Has severe, uncontrolled or active cardiovascular disorders. * Has serious or poorly controlled hypertension. * Has clinically significant bleeding symptoms or significant bleeding tendency within 1 month prior to the first dose. * Has untreated brain or Central nervous system (CNS) metastases or brain/CNS metastases that have progressed. * Has evidence of current ILD/non-infectious pneumonitis or a prior history of ILD/non-infectious pneumonitis requiring high-dose glucocorticoids Or suspected ILD/non-infectious pneumonitis that cannot be ruled out by imaging. * Has ongoing adverse reaction(s) from prior therapy that has(have) not recovered to Grade 1 or to the baseline status preceding prior therapy. * Has received any prior therapy with an Antibody-drug conjugate (ADC) with a Topoisomerase-1 (TOPO1)-inhibitor payload. * Is pregnant or breastfeeding.
