What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests how tirzepatide affects hot flashes and signs of how the body ages. It is for women ages 46–60 who have gone through menopause, have bothersome hot flashes, and meet the study’s weight rules.
The purpose of this study is to determine the effect of tirzepatide on vasomotor symptoms and on measures of biological aging.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- You are female and age 46–60.
- Your BMI, a measure of weight compared with height, is at least 30. A BMI of at least 27 also qualifies if you have high blood pressure, unhealthy blood fat levels, sleep apnea (breathing pauses during sleep), or heart or blood vessel disease.
- You average at least 28 hot flashes over 2 weeks, severe enough that you have sought treatment.
- You have had hot flashes for more than 30 days before joining.
- You are willing to give yourself shots of the study drug.
- You give informed consent, meaning you understand the study and agree to join.
- You can and will follow a personal eating plan with about 500 fewer calories than your body needs each day, but no less than 1,000 calories daily.
- You will do at least 150 minutes of moderate or harder exercise each week.
Who can't join
- You use, used in the past 4 weeks, or plan to use estrogen, including estrogen birth control or vaginal estrogen, androgens, or progestogens. You also cannot have used hormone therapy, talk therapy or hypnosis for menopause symptoms, fezolinetant, or elinzanetant in the past 4 weeks.
- Cancer treatment caused your menopause; your kidney filtering rate is 30 or less; or your thyroid tests show TSH of at least 7 with low free T4. Active inflammation, immune system disease, infection, liver or digestive disease, cancer, or uncontrolled mental illness also excludes you.
- Your weight changed by more than 5% in the past 3 months or by at least 20 pounds in the past 6 months, or you used another weight-loss drug in the past 3 months.
- You had or plan weight-loss surgery, with an exception for liposuction or a tummy tuck more than 1 year ago. Past or planned scope- or device-based treatment, or device removal within 6 months, also excludes you.
- You take drugs that can cause weight gain unless your dose and weight have been stable for at least 3 months. You also cannot have used ongoing steroid medicine that affects the whole body for more than 2 weeks in the past 3 months.
- You are pregnant, breastfeeding, planning pregnancy, or able to become pregnant without using highly effective birth control.
- You have a reason you cannot safely take tirzepatide, including a personal or family history of medullary thyroid cancer, multiple endocrine neoplasia type 2 (a condition that causes gland tumors), or an allergy to tirzepatide or its ingredients.
- You are in another study testing an experimental product, or received study treatment or placebo in one in the past 30 days. You also cannot have surgery planned that requires sedation or general anesthesia during this study or within 2 weeks after your last dose.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria 1. Females 2. Age 46-60 years old. 3. BMI ≥30 kg/m2 or BMI ≥27 kg/m2 in the presence of adiposity-associated diseases (hypertension, dyslipidemia, obstructive sleep apnea, cardiovascular disease). 4. Presence of bothersome hot flashes (an average of ≥ 28 episodes during a consecutive 2-week period with symptom severity sufficient to have prompted the participant to seek therapeutic treatment). 5. Hot flashes must be present for \>30 days prior to study entry. 6. Willingness to self-inject drug 7. Provided informed consent to be part of the study. 8. Willingness and capability to follow an individualized hypocaloric diet designed to achieve an approximate energy deficit of 500 kcal/day relative to estimated total daily energy requirements. Daily energy requirements will be calculated using Harris-Benedict equation to estimate resting energy expenditure and multiplied by a sedentary physical activity factor of 1.2. Prescribed daily caloric intake will not be less than 1,000 kcal/day. Participants must also be willing to engage in at least 150 minutes per week of moderate-intensity or greater physical activity. Exclusion Criteria 1. Current or recent use (past 4 weeks) treatment with menopausal hormone therapy. 2. Any current, recent use (past 4 weeks), or planned use of: 1. Estrogen-containing contraceptive methods or menopausal hormone therapy (oral, transdermal, high dose vaginal ring, injection, pellets). 2. Vaginal estrogen. 3. Androgens. 4. Progestogens. 3. Current or recent use (past 4 weeks) treatment for menopausal symptoms with cognitive behavioral therapy and/or hypnosis. 4. Current or recent use (past 4 weeks) of fezolinetant or elinzanetant. 5. Menopause as a result of cancer treatments. 6. Impaired renal function (GFR ≤30 ml/min/1.73 m²). 7. Thyroid-stimulating hormone ≥7 with low free T4. 8. Active inflammatory, autoimmune, infectious, hepatic, gastrointestinal, malignancy, or uncontrolled psychiatric disease. 9. \>5% change in weight during the 3 months prior to screening and, or eight fluctuation of ≥20 pounds within the past 6 months (self-report). 10. Other obesity medication was used within the past 3 months. 11. History of bariatric surgery. Prior or planned surgical treatment for obesity (excluding liposuction or abdominoplasty performed \> 1 year before screening). 12. Past or intended endoscopic and/or device-based therapy or removal within last six months. 13. Use of weight gain-promoting medications (including tricyclic antidepressants, atypical antipsychotics, and mood stabilizers), unless the participant has been receiving the medication at a stable dose for ≥3 months prior to enrollment and has maintained a stable body weight during that time 14. Current or recent (within 3 months) use of chronic systemic glucocorticoid therapy for over 2 weeks within the past 3 months. 16\. Female who is pregnant, breastfeeding, or intends to become pregnant or is of child-bearing potential and not using a highly effective contraceptive method. 15\.
