What to expect
No one can promise a trial will help you personally — that is what the research is trying to find out.
What participation involves
Not stated in the study description — ask the study team about visits and procedures.
- Compensation
- Not stated
- Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
- Participants are monitored closely by the research team throughout the study.
- Some participants may receive access to an investigational treatment, which is still being studied.
- Your participation helps researchers learn whether this approach could help future patients.
About this study
In plain language
This study tests ITI-1284 for people with Alzheimer's disease who have psychosis, such as seeing things that are not there or having false beliefs. It compares ITI-1284 with a placebo (a treatment with no active drug) to see how well it works and how safe it is.
This is a multicenter, randomized, double-blind, parallel-group, placebo-controlled, flexible-dose study evaluating the efficacy, safety, and tolerability of ITI-1284 compared with placebo in the treatment of psychosis in patients with AD.
Compensation is read from the study's own description and may be missing — always confirm with the study team.
Eligibility
Who can join
- Can give signed consent, or an authorized person can give consent for them. If able, the patient must also agree.
- Has Alzheimer's disease confirmed by a doctor's assessment and a blood test, spinal fluid test, or special brain scan.
- Has psychosis, such as false beliefs or seeing or hearing things that are not there, at both starting visits.
- Scores at least 2 on at least one question about false beliefs or seeing or hearing things on the BEHAVE-AD symptom scale at both starting visits.
- Scores at least 4 on the CGI-S scale, which rates how severe the illness is, at both starting visits.
- Scores 6 to 24 on the MMSE-2 memory and thinking test. Can understand and answer spoken questions and follow study steps.
- Has a caregiver, such as a family member, friend, housemate, or paid caregiver.
Who can't join
- Has psychosis mainly caused by sudden confusion, substance abuse, or another medical condition rather than Alzheimer's disease.
- Has schizophrenia or another psychotic disorder not related to Alzheimer's disease, or bipolar disorder.
- Is at risk of suicide or is an immediate danger to themselves or others, or has attempted suicide in the past 2 years.
- Cannot or will not stop certain medicines that affect mood, thinking, or the brain and nerves, as reviewed by the study sponsor.
- Is in the hospital or receiving skilled nursing care for a condition other than dementia.
- Is confined to bed, or has a serious, unstable medical condition that could make the study unsafe or make results hard to judge.
- Is receiving hospice or end-of-life care.
Simplified for readability. The official criteria below are what the study team uses.
Official eligibility criteria
Inclusion Criteria: 1. Can understand the nature of the trial and protocol requirements and provide signed informed consent, if in the judgement of the Investigator is deemed competent to provide consent or if patient is deemed not competent to provide informed consent, with the patient's assent (if capable), consent may be provided by an appropriate person (eg, patient's Legally Authorized Representative \[LAR\]) before the initiation of any study-specific procedures in accordance with local regulations 2. Meets clinical criteria for AD based on 2011 NIA-AA criteria and either: 1. Has a high likelihood for amyloid pathology consistent with AD, as confirmed by blood-based biomarker at Screening as detected by an appropriately validated test; or 2. Has documented confirmation of AD by cerebrospinal fluid (CSF) biomarker or amyloid positron emission tomography (PET) brain scan 3. Meets criteria for psychosis in accordance with the International Psychogeriatric Association (IPA) provisional consensus definition at Screening and Baseline 4. Scoring ≥ 2 on any item of the BEHAVE-AD Part A. Paranoid and Delusional Ideation item and/or Part B. Hallucinations item (ie, psychosis subscale) at Screening and Baseline 5. CGI-S score ≥ 4 at Screening and Baseline 6. Mini-Mental State Examination, 2nd Edition™: Standard Version (MMSE-2®:SV) score of 6 to 24 (inclusive) at Screening with sufficient verbal ability to understand and answer questions and comply with procedures 7. Has a designated caregiver (eg, relative, housemate, close personal friend, or professional caregiver) Exclusion Criteria: 1. Psychotic symptoms that are primarily attributable to delirium, substance abuse, or another general-medical condition (for example, hypothyroidism) or has been diagnosed with one or more of the following psychiatric conditions: 1. Schizophrenia, schizoaffective disorder, or other psychotic disorder that is not related to Alzheimer's dementia 2. Bipolar disorder 2. Risk for suicidal behavior during the course of their participation in the study or is considered to be an imminent danger to themselves or others, in the opinion of the investigator, and/or as assessed by C-SSRS; or the patient has had 1 or more suicide attempts within 2 years prior to Screening 3. The patient is unable or unwilling to discontinue other drugs with known psychotropic properties or any non-psychotropic drugs with known or potentially significant central nervous system effects, as reviewed by the Sponsor or designee 4. The patient is hospitalized or receiving skilled nursing care for any medical condition other than dementia 5. The patient is bedridden or has any significant medical condition that is unstable and would either: 1. Place the patient at undue risk from study drug or undergoing study procedures; or 2. Interfere with the interpretation of safety or efficacy evaluations performed during the course of the study 6. The patient is in hospice or end-of-life care
