What to expect

No one can promise a trial will help you personally — that is what the research is trying to find out.

What participation involves

Not stated in the study description — ask the study team about visits and procedures.

Compensation
Not stated
  • Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
  • Participants are monitored closely by the research team throughout the study.

About this study

In plain language

This study tests repotrectinib for solid tumors that have grown nearby or spread and have certain gene changes. Phase 1 checks safe doses in adults with ALK, ROS1, or NTRK gene changes. Phase 2 checks how well it works in people age 12 and older with ROS1 or NTRK gene changes.

Phase 1 dose escalation will determine the first cycle dose-limiting toxicities (DLTs), the maximum tolerated dose (MTD), the biologically effective dose and recommended Phase 2 dose (RP2D) of repotrectinib given to adult subjects with advanced solid malignancies harboring an ALK, ROS1, NTRK1, NTRK2, or NTRK3 gene rearrangement. Midazolam DDI substudy will examine effect of of repotrectinib on CYP3A induction. Phase 2 will determine the confirmed Overall Response Rate (ORR) as assessed by Blinded Independent Central Review (BICR) of repotrectinib in each subject population expansion cohort of advanced solid tumors that harbor a ROS1, NTRK1, NTRK2, or NTRK3 gene rearrangement. The secondary objective will include the duration of response (DOR), time to response (TTR), progression-free survival (PFS), overall survival (OS) and clinical benefit rate (CBR) of repotrectinib in each expansion cohort of advanced solid tumors that harbor a ROS1, NTRK1, NTRK2, or NTRK3 gene rearrangement.

Compensation is read from the study's own description and may be missing — always confirm with the study team.

Eligibility

Who can join

  • Have an advanced solid tumor with a gene change confirmed by testing: ALK, ROS1, or NTRK1–3 for Phase 1; ROS1 or NTRK1–3 for Phase 2.
  • Be at least 18 for Phase 1 or 12 for Phase 2. Local rules may require age 20 or older.
  • Be fully active or able to do light work, and be expected to live at least 3 months.
  • Have at least one tumor that can be measured. Tumors only in the brain or spinal cord may qualify. Spread to these areas or their lining without symptoms may also qualify under study rules.
  • Meet study limits for blood counts, kidney and liver function, and blood minerals.
  • For Phase 1, be able to swallow whole capsules. Side effects from past cancer treatment must be mild or gone, except hair loss. Past chemotherapy or immune treatment is allowed.
  • For Phase 2, provide tumor tissue for gene testing and meet rules about past treatment for your study group.
  • For Phase 2, provide written consent. People ages 12–17 need a parent or legal guardian to sign.

Who can't join

  • Taking part in another clinical trial that tests a treatment.
  • Cancer spread to the brain or the lining of the brain or spinal cord that causes symptoms.
  • Another cancer that needed treatment in the past 2 years, except certain skin cancers or fully removed cancers that had not spread beyond where they started.
  • Major surgery within 4 weeks or radiation within 2 weeks before starting the study. Short courses of radiation for bone pain may be allowed if finished at least 48 hours before entry.
  • Certain serious heart or blood vessel problems now or in the past 6 months, or heart rhythm problems or risk factors listed in the study rules.
  • A known active infection, including HIV, or a gut condition that affects how your body absorbs medicine.
  • Nerve damage outside the brain and spinal cord that is moderate or worse.
  • Certain serious lung scarring or lung inflammation, now or in the past. Past lung inflammation from radiation alone does not rule you out.

Simplified for readability. The official criteria below are what the study team uses.
