What to expect

No one can promise a trial will help you personally — that is what the research is trying to find out.

What participation involves

Not stated in the study description — ask the study team about visits and procedures.

Compensation
Not stated
  • Study-related exams and procedures are typically provided at no cost. This is how many trials work in general — confirm the details for this study with the study team.
  • Participants are monitored closely by the research team throughout the study.
  • Some participants may receive access to an investigational treatment, which is still being studied.
  • Your participation helps researchers learn whether this approach could help future patients.

About this study

In plain language

This study tests how safe rezatapopt (PC14586) is and how well it works for solid tumors that have grown nearby or spread and have a TP53 Y220C gene change. Phase 2 is now enrolling and tests rezatapopt alone. Phase 1 tests different doses alone, and Phase 1b tests it with pembrolizumab.

The Phase 2 monotherapy portion of this study is currently enrolling and will evaluate the efficacy and safety of PC14586 (INN rezatapopt) in participants with locally advanced or metastatic solid tumors harboring a TP53 Y220C mutation. The Phase 1 portion of the study will assess the safety, tolerability and preliminary efficacy of multiple dose levels of rezatapopt as monotherapy and in Phase 1b in combination with pembrolizumab.

Compensation is read from the study's own description and may be missing — always confirm with the study team.

Eligibility

Who can join

  • You are 18 or older, or age 12–17 with approval from the study’s safety review group.
  • Your solid tumor has grown nearby or spread and has a TP53 Y220C gene change.
  • You can do all or most daily activities (ECOG score of 0 or 1).
  • Your cancer has gotten worse after at least one type of cancer treatment.
  • Your organs work well enough for the study.
  • For Phase 2, your cancer can be measured using the study’s rules.
  • For Phase 1b, your cancer can be measured.
  • For Phase 1b, you have never had anti-PD-1/PD-L1 treatment, which helps the immune system fight cancer, or your cancer got worse during it.

Who can't join

  • You had cancer treatment within 21 days (or 5 drug half-lives—the time it takes your body to clear most of a drug), radiation within 14 days, or certain medicines that strongly affect liver enzymes CYP3A4 or CYP2C9 within 14 days before the study drug.
  • You have a tumor that started in the brain or spinal cord; a history of cancer in their lining or pressure on the spinal cord; or cancer that spread to the brain unless nerve-related symptoms are stable without steroids. A stroke or mini-stroke within 6 months also excludes you.
  • You have certain heart problems: recent unstable chest pain or heart attack within 6 months, uncontrolled high blood pressure, heart failure, or abnormal heart rhythms.
  • You have a digestive illness that may affect how you absorb the drug, cannot take medicine by mouth, or have had an organ transplant.
  • You have another known active cancer, except treated abnormal cervical cells or non-melanoma skin cancer; or active, uncontrolled hepatitis B, hepatitis C, or HIV infection.
  • For Phase 2, you have a KRAS gene change called a single nucleotide variant, unless you have ovarian cancer.
  • For Phase 1b, you previously took rezatapopt; stopped certain immune treatments because of a severe immune side effect; had a severe allergy to pembrolizumab or its ingredients; or had a live vaccine within 30 days.
  • For Phase 1b, you have a weak immune system, ongoing whole-body steroid use within 7 days, an immune disease needing whole-body treatment in the past 2 years, past lung swelling from radiation, noninfectious lung swelling or scarring needing steroids, an active infection needing whole-body treatment, or any history of HIV.

Simplified for readability. The official criteria below are what the study team uses.

Official eligibility criteria

Inclusion Criteria: * At least 18 years of age or 12 to 17 years of age after Safety Review Committee approval. * Locally advanced or metastatic solid malignancy with a TP53 Y220C mutation * Eastern Cooperative Oncology Group (ECOG) status of 0 or 1 * Previously treated with one or more lines of anticancer therapy and progressive disease * Adequate organ function * Measurable disease per RECIST v1.1 (Phase 2) Additional Criteria for Inclusion in Phase 1b (rezatapopt) + pembrolizumab combination) * Anti-PD-1/PD-L1 naive or must have progressed on treatment * Measurable disease Exclusion Criteria: * Anti-cancer therapy within 21 days (or 5 half-lives) of receiving the study drug * Radiotherapy within 14 days of receiving the study drug * Primary CNS tumor * History of leptomeningeal disease or spinal cord compression * Brain metastases, unless neurologically stable and do not require steroids to treat associated neurological symptoms * Stroke or transient ischemic attack within 6 months prior to screening * Heart conditions such as unstable angina within 6 months prior to screening, uncontrolled hypertension, a heart attack within 6 months prior to screening, congestive heart failure, prolongation of QT interval, or other rhythm abnormalities * Strong CYP3A4 inducers and strong CYP2C9 inhibitors/inducers within 14 days of first dose of rezatapopt * History of gastrointestinal (GI) disease that may interfere with absorption of study drug or patients unable to take oral medication * History of prior organ transplant * Known, active malignancy, except for treated cervical intraepithelial neoplasia, or non-melanoma skin cancer * Known, active uncontrolled Hepatitis B, Hepatitis C, or human immunodeficiency virus infection Additional Criteria for Exclusion from Phase 2 (rezatapopt monotherapy) * Known KRAS mutation, defined as a single nucleotide variant (SNV) (Phase 2).
